Abstract

Sirs: Evidence on the influence of apolipoprotein E (APOE) polymorphisms on multiple sclerosis (MS) disease severity has been controversial. Several studies have associated the ε4 polymorphism with more rapid progression [1, 2, 7]. Others have not confirmed this [5, 6, 10, 11]. The MS severity score (MSSS) was recently developed as a powerful method for comparing disease progression in genetic association studies [4]. In the present study we investigated the effect of APOE genotypes on disease severity in Greek MS patients, a previously unstudied population, using the MSSS.APOE genotypes were determined in 212 Greek patients (135 females, 77 males) with MS (McDonald criteria) under the care of the Neurology Department, University of Athens [3]. Only patients with disease duration of at least one year were included. Mean patient age was 38.7 ± 10.4 years (range, 16–69), mean age at onset 30.1 ± 9.9 years (range, 10–64), mean disease duration 8.6 ± 6.7 years (range, 1–39), mean EDSS...
