Abstract
We identified fatty acid desaturase fadslike and elongase elovllike genes from nibe croaker to better understand the molecular basis of n3 highly unsaturated fatty acid metabolism in marine fish Phylogenetic analysis revealed that the fadslike and elovllike genes were classified into the fads6 and elovl5 groups respectively We investigated the effects of various levels of docosahexaenoic acid DHAenriched live feed Artemia nauplii on larval growth survival and fadslike and elovllike gene expression After a 15day feed trial total length body weight and survival were all significantly improved by the supplementation of Artemia with DHA This result indicates that nibe croaker cannot endogenously produce enough DHA Furthermore the fadslike gene transcripts in larvae fed on oleic acidenriched Artemia were significantly higher than those in larvae on 100 DHAenriched Artemia In contrast no significant differences were observed in the transcript levels of the elovllike gene These data indicate that the fads6like gene was controlled by negative feedback from the quantity of DHA stored in the larval body These results have implications for the functionality of the fadslike gene in nibe croakerWe thank T Hirota Taiyo Yushi KK for the generous gift of DHA70G We also thank K Higuchi T Yatabe and Dr A Kurihara Tokyo University of Marine Science and Technology for experimental support and Dr JA Luckenbach Northwest Fisheries Science Center NOAA for reviewing the manuscript We are grateful to members of the Tateyama field station J Ito M Masuko N Ishikawa S Ube and N Hayashi for their assistance in the maintenance of the fish
