Abstract
The steroidal alkaloid 3β 20αdiaminopregn5ene kurchamine obtained from the stem bark of Holarrhena antidysenterica is reported to have appreciable amoebicidal activity Its three stereoisomers namely 3α 20βdiaminopregn5ene 3β 20βdiaminopregn5ene and 3α20αdiaminopregn5ene and their intermediate stereoisomeric monoamino pregnene alkaloids namely 3βaminopregn5ene20one 3αaminopregn5ene20one 20α aminopregn5ene3βol 20βaminopregn5ene3βol were synthesized The natural stereoisomer and synthesized diamino and monoamino stereoisomers were Nmethylated and all the compounds were evaluated for amoebicidal activity comparison The natural stereoisomer 3β20αdiaminopregn5ene kurchamine was found to be superior than other stereoisomers and Nmethylation was found to have insignificant effect on amoebicidal activity of stereoisomersStereoisomers of kurchamine the natural pregnene alkaloid of Holarrhena antidysenterica stem bark and their intermediate monoamino stereoisomers were synthesized from 16dehydropregnenolone acetate 16DPA Kurchamine and synthesized stereoisomers along with their Nmethylated analogs were evaluated for their antiamoebic activityThe authors are thankful to Dr G L Sharma and Ravinder Kour of Organic Chemistry Division Regional Research Laboratory Jammu RRL for biological testing of the compounds RMV is thankful to the Council of Scientific and Industrial Research CSIR for the award of Junior Research Fellowship JRF
