Abstract
We describe expression and characterization of recombinant human colorectal cancer antigen GA7332 fused to Fc fragment of antibody GA7332Fc using a replicating vector based on Beet curly top virus in infiltrated Nicotiana benthamiana leaves Recombinant GA7332Fc/KDEL with a molecular mass of ~68 kDa was transiently expressed The level of expression of GA7332Fc with ER retention signal KDEL GA7332Fc/KDEL in the expression vector system was 096 of total soluble proteins Recombinant GA7332Fc/KDEL was purified using an affinity chromatography Mice immunized with recombinant GA7332Fc/KDEL mounted a strong GA7332Fc/KDELspecific serum antibody response Vaccination of plantderived recombinant GA7332Fc/KDEL regressed tumor volumes in BALB/c mice The population of activatedT and NKT cells increased notably in lymph node spleen and tumorinfiltrating lymphocytes derived from the tumorregressed mice Taken together recombinant GA7332Fc/KDEL expressed in plants can be used as an effective experimental immunogen for research in cancer vaccine developmentThis research was supported by a grant from the Basic Science Research Program through the National Research Foundation of Korea NRF funded by the Ministry of Education Science and Technology 20110003112 and a Grant 111096031HD110 from Korea Institute of Planning and Evaluation for Technology in Food Agriculture Forestry and Fisheries
