Effect of trophic conditions and humic acid on ala

Abstract

In dialyzed extracts from winter wheat plants an intensive enzymatical transamination reaction occurred between Lαalanine and αketoglutaric acid Lαalanine + + 2oxoglutarate = pyruvate + Lglutamate EC 2612 as well as a weak nonenzymatical transamination reaction practically immeasurable Pyridoxal5’phosphate strongly affected the reaction rate Besides the transamination productglutamate γaminobutyric acid was formed in this reaction This amino acid could have originated neither via proteolysis of the enzyme extract nor via decarboxylation from glutamate formed nor via transamination of succinicγsemialdehyde after αketoglutarate decarboxylation This was the only case of its formation in the transamination reactions investigated in our laboratory — it originated from the alanineglutamate reaction only and the mechanism of its biosynthesis cannot be elucidated for the presentThe effect of trophic conditions Knop’s nutrient solution a water solution of potassium humate water was not revealed when calculating per dry weight unit However when calculating per protein unit an increased activity was found in the dialysates from shoots of both nutrient — deficient variants Roots of plants cultivated in potassium humate had the lowest activityThe discussion concerns the possibility of an adaptive use of this transamination for increasing the essential glutamate level in green parts of the plants cultivated under unfavourable nutritive conditions and also deals with a further characteristic of the differing metabolism of plants cultivated in humateV dialyzovaných extraktech z rostlin ozimé pšenice probhá intenzívní transaminační enzymová reakce mezi Lαalaninem a kyselinou aketoglutarovou Lαalanin + 2oxoglutarát = = pyruvát + Lglutamát EC 2612 a slabá prakticky neměřitelná transaminační reakce neenzymová Pyridoxal5′fosfát výrazně ovlivnil aktivitu reakce Vedle transaminačního produktuglutamátu se při reakci vytvářela kyselina γaminomáselná Tato aminokyselina nemohla vzniknout ani proteolyticky z enzymového extraktu ani dekarboxylačně ze vznikajícího glutamátu ani transaminací semialdehydu kyseliny jantarové po dekarboxylaci αketoglutarátu Její vznik byl zatím v našich transaminačních reakcích ojedinělý — vyakytla se pouze p0159i reakci alaninglutamát a mechanismus její biosyntézy zatím nemohl být objasněnVliv trofickýoh podmínek Knopův živný roztok vodný roztok kaliumhumátu voda se při přepočtu na sušinu neprojevil Při přepočtu na bílkoviny měly vyšší aktivitu dialyzáty z nadzemních čcástí obou troflcky deácitních variant kaliumhumát voda Kořeny varianty kaliumhumátu měly nejnižší aktivituJe diskutována možnost adaptivního využití této transaminace k zvyšování nezbytné hladiny glutamátu v zelených částech rostlin pěstovaných v nepříznivých trofických podmínkách a zároveň i další rys odlišného metabolismu rostlin pěstovaných ve variantě s kyselinou huminovou
