Abstract
A sensitive and rapid method for simultaneous determination of baicalin baicalein wogonoside wogonin berberine copstisine and palmatine of Scutellaria baicalensis and Coptidis rhizoma extract in rat plasma based on ultra highperformance liquid chromatography coupled with tandem mass spectrometry UPLC–MS/MS was developed Plasma samples were processed by protein precipitation with methanol Chromatographic analysis was performed on an Acquity BEH C18 column 100 mm × 21 mm 17 µm using a mobile phase consisting of acetonitrile and water containing 01 formic acid Electrospray ionization in positive ion mode and multiple reaction monitoring were used to identify and quantify active components All calibration curves gave good linearity r 0991 over the concentration range from 072 to 375 ng/mL for all components Recovery was 5811–10283 depending on the analyte and interday variability was less than 13 The lower limit of quantification for all compounds was between 082 and 133 ng/mL The intra and interday precision and accuracy of the quality control samples at three concentrations showed ≤147 relative standard deviation and 866–1055 accuracy The validated method was successfully applied to determine the concentrations of seven bioactive components in rat plasma after oral administration of Radix scutellariae and Coptidis rhizoma extractThis work was financially supported by National Basic Research Program of China 973 Program 2011CB505300 2011CB505303 the Nature Science Foundation of China No 81072996 81102743 Jiangsu Collaborative Innovation Center of Chinese Medicinal Resources Industrialization A Project Funded by the Priority Academic Program Development of Jiangsu Higher Education Institutions PAPD Construction Project for Jiangsu Key Laboratory for High Technology of TCM Formulae Research BM2010576
