Abstract
Sulfolobus acidocaldarius a hyperthermoacidophilic archaeon possesses two βdecarboxylating dehydrogenase genes saci 0600 and saci 2375 in its genome which suggests that it uses these enzymes for three similar reactions in lysine biosynthesis through 2aminoadipate leucine biosynthesis and the tricarboxylic acid cycle To elucidate their roles these two genes were expressed in Escherichia coli in the present study and their gene products were characterized Saci 0600 recognized 3isopropylmalate as a substrate but exhibited slight and no activity for homoisocitrate and isocitrate respectively Saci 2375 exhibited distinct and similar activities for isocitrate and homoisocitrate but no detectable activity for 3isopropylmalate These results suggest that Saci 0600 is a 3isopropylmalate dehydrogenase for leucine biosynthesis and Saci 2375 is a dual function enzyme serving as isocitratehomoisocitrate dehydrogenase The crystal structure of Saci 0600 was determined as a closedform complex that binds 3isopropylmalate and Mg2+ thereby revealing the structural basis for the extreme thermostability and noveltype recognition of the 3isopropyl moiety of the substrateThis work was supported in part by JSPS KAKENHI Grant No 24228001 MN We are grateful to the staff of the Photon Factory for their assistance with data collection which was approved by the Photon Factory Program Advisory Committee Proposal nos 2011G524 2013G618 KL was supported by an ERC starting grant ARCHAELLUM 311523 and intramural funds from the Max Planck Society
