Abstract
A homogenous lightinduced chemiluminescence immunoassay AlphaLISA method was established for the determination of residues of zearalanol and its analog zearalanone in muscle tissue samples AlphaLISA is a beadbased proximity assay When donor and acceptor beads proximity a cascade of chemical reactions begin The end result is a greatly amplified signal that contributes to the detection sensitivity down to the attomole level Compared with other methods the AlphaLISA has characteristics of homogeneity being free of cleaning high sensitivity The method showed a linear relationship in the range of 001–4 ng/mL R 2 099 the sensitivity of the assay was 0066 ng/mL Crossreactivity rate of zearalanol was 100 and zearalanone was 821 and other compounds were not more than 40 The average recovery rates of Zearalanol at spiked levels of 1–4 ng/mL were 963 to 1050 and 917 to 1005 for pork and bovine muscles the intraday precision ranged from 26 to 98 and the interday precision ranged from 87 to 175 These results indicated that the proposed method was successfully applied in the analysis of zearalanol and its analog zearalanone in muscle tissuesThis work was financially supported by “Chinese National Inspection and Quarantine Nonprofit Industry Research No 201210086” and “General Administration of Quality Supervision Inspection and Quarantine Science and Technology Project No 2015IK333”Qian Gong declares that she has no conflict of interest Cunxian Xi declares that he has no conflict of interest Fuping Nie declares that she has no conflict of interest Shurui Cao declares that she has no conflict of interest Bobing Tang declares that he has no conflict of interest Lei Zhang declares that he has no conflict of interest Xianliang Li declares that he has no conflict of interest Guoming Wang declares that she has no conflict of interest Dongdong Chen declares that she has no conflict of interest Zhaode Mu declares that she has no conflict of interest
