Abstract
Expressions of HIF2α type X collagen COL10 matrix metalloproteinase MMP13 and vascular endothelial growth factor VEGF in nonOA and OA menisci were analyzed by realtime RTPCR and immunohistochemistry IHC Meniscal cells from OA patients were treated with interleukin1β IL1β and gene expression was analyzed After knockdown of HIF2α in OA meniscal cells COL10 and MMP13 expression were analyzed by RTPCR western blotting immunofluorescence and ELISAHistological analysis demonstrated weak staining of the superficial layer and large round cells in OA meniscus RTPCR analysis showed that HIF2α COL10 MMP13 and VEGF mRNA expressions were higher in OA than nonOA meniscal cells IHC showed a coordinated staining pattern of HIF2α COL10 and MMP13 in OA meniscus IL1β treatment increased HIF2α COL10 and MMP13 expressions in OA meniscal cells and knockdown of HIF2α suppressed IL1βmediated increase in COL10 and MMP13 expression
