Abstract
Total colonyforming units CFU were more sensitive to fludarabine than to bendamustine in agar colony assays IC50 07 μM/L and 85 μM/L respectively Using the Bliss independence model and combining the two agents yielded additive inhibition of progenitors Nonclonogenic assays including LTC and an apoptosis assay detecting activated caspases showed that stem cells are characterized by low sensitivity to bendamustine In contrast fludarabine strongly inhibited the viability and growth of stem cells in LTCWe thank Ms K Freiberg D Freese and C TietzeBürger for their excellent technical assistance Dr Weirowski made valuable grammatical and stylistic suggestions All authors have critically read and approved the manuscript MS AB WK ET and IWB design of the study BR LTC and agar colony experiments MS and AB apoptosis experiments MS has written the manuscript ET head of the department The project has been financially supported by Ribosepharm GmbH Gräfelfing and Mundipharma Limburg Germany
