Abstract

Helicase involves many cellular processes that separate doublestranded nucleic acid into single strands Although it is believed that helicase translocates nucleic acids it is difficult to show the direct evidence of translocation on nucleic acids In this study an avidinbiotinylated nucleotidesbased method for helicase translocation assay has been described and the biochemical assay results have been demonstratedThe author would like to thank Dr Zvi Kelman for his input during the review of this manuscript Dr Jerard Hurwitz for providing SpMCM4/6/7 and SV40 large Tantigen Dr Mike O’Donnell for E coli helicases and Dr Gyri Haugland for the Thermoplasma acidophilum MCM This research has been supported by Kyungpook National University Research Fund 2007
